FOXO4-DRI is an all-D-amino-acid, retro-inverso peptide derived from a segment of the transcription factor Forkhead box O4 (FOXO4), fused to a cell-penetrating import motif. It was described by de Keizer and colleagues as a tool for disrupting the FOXO4–p53 interaction that sequesters p53 in the nuclei of senescent cells.

The retro-inverso design — reversed sequence assembled from D-amino acids — is used to improve proteolytic stability relative to the native L-peptide while preserving the intended interaction surface.
